This study aims to analyze the need for the development of a biochemistry practicum on DNA isolation material. The method used was a descriptive method with a quantitative approach. The subjects involved in the study consisted of three biochemistry lecturers, 29 students who had received biochemistry lectures, and eight alumni who worked as chemistry teachers in high schools. Data were collected using non-test instruments, namely through curriculum document analysis, RPS (semester lesson plan), questionnaires, and interviews. The results obtained indicate that DNA isolation material has the potential to be developed in a biochemistry practicum. Practicum development is based on the results of document analysis that has been done, stating that DNA isolation is crucial to be taught to students. The results of student perceptions require a variety of learning methods and the utilization of facilities, and with DNA isolation, this can be maximized. © 2025 Author(s).
